The Department of Biophysics and Neuroscience was established in 2000 by the merger of two scientific organisational units of the Faculty of Medicine: the Department of Biophysics and the Independent Neuro System Biophysics Laboratory.

The Head of the Department is Prof. Jerzy Mozrzymas.

The scientific activities of the Department of Biophysics and Neuroscience focus on:

  1. pharmacology of GABAergic inhibitory transmission in the central nervous system,

  2. effects of structural changes at the single amino acid level on GABAA receptor function,

  3. molecular and cellular mechanisms of synaptic plasticity phenomena in the brain, with particular emphasis on the plasticity of inhibitory transmission,

  4. evaluation of mechanisms of action and anticancer activity of new synthetic compounds and compounds of natural origin,

  5. the problem of multidrug resistance of tumours to cytotoxic drugs,

  6. effects of natural substances and selected drugs on model lipid membranes of different compositions,

  7. QSAR study of the effect of substituent effects on the structure and properties of chemical molecules,

  8. assessment of interactions within the gut-brain axis and the influence of intestinal dysbiosis on drug activity,

  9. studies on pharmacological modulation of voltage-gated potassium channel activity in normal and cancer cells,

  10. multidrug resistance of tumours to cytotoxic drugs and resistance of pathogenic fungi to antifungal drugs.

The Department of Biophysics and Neuroscience has extensive research and didactic facilities. The laboratory classes for students are held in the building of the Department at ul. Mikulicza-Radeckiego 9.

Laboratory of electrophysiological measurements in brain slices

The workstation is equipped with a Leica 1200S vibratome for the preparation of brain sections. With this device, sections of the mouse brain are prepared in cooled artificial cerebrospinal fluid and used for electrophysiological studies afterwards.

A workstation for patch clamp electrophysiological tests in brain sections, equipped with: a Nikon Eclipse FN1 fluorescence microscope with differential interference contrast, two motorised PatchStar micromanipulators, Digidata1550 acquisition system and a Multiclamp 700B amplifier. The set enables measurement of single synapse activity at specific neuronal projections in brain tissue. Additionally, the workstation is also equipped with a Picospritzer, which can be used to apply small amounts of a test substance directly to living tissue and examine its effects using electrophysiological techniques.

A workstation for patch clamp electrophysiological tests in brain sections, equipped with: Zeiss AxioExaminer Z1 fluorescence microscope with differential interference contrast, two motorised PatchStar micromanipulators, Digidata1550 acquisition system and a Multiclamp 700B amplifier. Additionally, the microscope is equipped with a Sutter DG-4 fluorescent light source, which can be controlled with submillisecond precision, which enables the stimulation of selected, defined neurons using short light pulses. The workstation is also equipped with a 445nm laser coupled with the UGA-42 Firefly galvanometer - based system for photomanipulation. This device uses a laser neurotransmitter to manipulate selected and identified synapses of a specific neuron. The workstation gives unique opportunities to study the activity and plasticity of synapses.

A workstation for measuring field potentials in brain sections equipped with: Leica microscope with differential interference contrast, two MX110 Siskiyou manual manipulators and a Digidata 1440 acquisition system. There are two such workstations in the Laboratory.

Single-channel workstation

"Theta 3" electrophysiological test workstation. The workstation is used to measure single-channel recordings using the patch-clamp technique in a cell-attached configuration. The recordings from the individual channels are current waveforms of very low amplitude (approx. 2pA), so the workstation is adapted to isolate external network noise as much as possible. For that purpose, it was equipped with a sealed Faraday cage located on an anti-vibration table, which is closed during recording. Inside the cage, there is an inverted Leica microscope (Dmi8) equipped with a camera and a fluorescence illuminator (470nm, CoolLED) and a head with a measuring electrode controlled from outside the cage by a micromanipulator (Scientifica PatchStar). Patch-clamp recordings are made using the Axopatch 200B amplifier and the Digidata 1550B acquisition system (Molecular Devices) with the appropriate pClamp software (Molecular Devices) controlled from a desktop computer located at the workstation. Necessary tools for the single-channel recording measurement with noise reduction are properly prepared pipettes placed on the electrode and filled with a solution containing the selected concentration of the agonist. To prepare the pipettes, there is a glass pipette extractor (Sutter P-1000) at the workstation. Using a binocular, the area around the pipette tip is covered with thermosetting elastomer Sylgard (Dow Corning). There is also a micro-polishing device at the workstation, thanks to which it is possible to harden Sylgard (ejection of warm air) and to polish the tip of the pipette (approaching the heated filament) for optimal cell membrane-pipette contact.

Patch-clamp workstation for pharmacology and kinetics

"Theta 1" electrophysiological test workstation. This workstation is used to measure macroscopic currents using the patch-clamp technique in both whole cell (lifted cell) and membrane patch (excised patch) configurations. Two perfusion systems used at the workstation - an ultra-fast one based on a piezoelectric amplifier (PI, model HVPZT) with an infusion pump (WPI SP220i) and a fast BioLogic system (RSC 200) with an electro-valve system (EVH - 9) - enable conducting research using a wide range of concentrations of different agonists and pharmacological agents or compounds of natural origin. Additionally, the station is equipped with an anti-vibration table, Faraday cage, desktop computer, Nikon microscope (TS100) with fluorescence, Siskiyou micromanipulator (MC1000e-1), Digidata 1440A acquisition system (Molecular Devices) and Axopatch 200B amplifier (Molecular Devices).

This workstation is used to measure macroscopic currents using the patch-clamp technique in both whole cell (lifted cell) and membrane patch (excised patch) configurations. The ultra-fast perfusion system used at the workstation, based on a piezoelectric amplifier (PI, HVPZT model) with an infusion pump (Chemyx, Fusion 200), enables testing using a wide range of concentrations of different agonists and pharmacological agents or compounds of natural origin. Patch-clamp testing workstation in the building at ul. Mikulicza-Radeckiego 9

The patch-clamp testing workstation in the Laboratory at ul. Mikulicza-Radeckiego 9 conducts research in the field of modulation of voltage-gated Kv1.3-type potassium channels found in normal and cancer cells. The normal cells used for testing are T-lymphocytes isolated from the peripheral blood of healthy donors. The cancer cells are lymphoblastic cells of acute lymphocytic leukaemia - Jurkat T. The purpose of the research is to determine the effect of selected biologically active plant-derived polycyclic compounds with anticancer properties on the activity of Kv1.3 channels in cancer cells and, comparatively, in normal cells. The studied compounds belong to the group of: flavonoids, chalcones, their natural (prenyl) and synthetic (methoxyl) derivatives and statins.

Microcalorimetry Laboratory

The Microcalorimetry Laboratory of the Department of Biophysics and Neuroscience is equipped with two differential scanning calorimeters (Unipan and Rigaku). Differential scanning calorimetry (DSC) allows heat changes accompanying physical processes in macromolecule systems, such as denaturation or phase transitions, to be tracked. We mainly conduct research on phase transitions of model lipid membranes with different chemical compositions. Experiments on the effects of low-molecular-weight chemical compounds (e.g. medicines, naturally derived compounds and their derivatives) on the thermotropic properties of structures formed by hydrated phospholipids (multilayers or bilayers) make it possible to conclude on the effects of the test substances on the lipid structure of the cell membrane.

Molecular Biology Laboratory

The Molecular Biology Laboratory houses:

Workstations for the manipulation of cancer, fungal and bacterial cell lines enabling their multiplication (incubators, shaker-incubators), as well as work in sterile conditions (laminar chambers). The Laboratory is equipped with an automated cell counter that enables rapid assessment of the population of live and dead cells of various types and a wide range of sizes.

The workstations are equipped with thermocyclers, used for DNA amplification in the PCR reaction, reverse transcription and mutagenesis, nucleic acid electrophoresis apparatus and protein electrophoresis and transfer, used for separation of individual fractions. The Laboratory is also equipped with a UV chemiluminescence and UV fluorescence system by UVITEC Cambridge for visualising proteins and nucleic acids on blots and gels.

The workstations are equipped with microplate readers (Tecan Infinitie and BMG Labtech CLARIOstar) to measure absorption and fluorescence, used e.g. for cytotoxicity tests and measurement of protein activity. Additionally, the BMG Labtech CLARIOstar device is equipped with an electronic Atmospheric Control Unit (ACU) that enables independent control of the concentration of oxygen (O2) and carbon dioxide (CO2). In combination with temperature control and shaking, this allows cell tests to be performed under near-physiological conditions. The device offers the possibility of long kinetic measurements.

Laboratory of Bioinformatics

The Laboratory of Bioinformatics conducts research on the structure-function relationship of proteins. Protein sequence analysis software is used: T-Coffe, ClustalW, MUSCLE and Jalview. VMD and PyMol are used to visualise protein structures. Molecular dynamics simulations are carried out in the NAMD and GROMACS packages. The AutoDock Vind and HADDOCK software are used to simulate ligand docking. The PROPKA software is used to estimate the pKa values of the amino acid residues.

The Department of Biophysics and Neuroscience was established in 2020 by the merger of two scientific organisational units of the Faculty of Medicine: the Department of Biophysics and the Independent Neuro System Biophysics Laboratory.

The current head of the Department is Prof. Jerzy Mozrzymas.

The Division of Physics at the Medical Academy was established in 1952, the Division of Biophysics in 1967 and the Department of Biophysics in the 1980s. The position of the Head of the Division of Biophysics was successively held by: Prof. Andrzej Morawiecki (1967-1970), Prof. Marian Kochman (1971-1974), Prof. Stanisław Miękisz (1975-1997), Prof. Krystyna Michalak (1997-2019). The Independent Neuro System Biophysics Laboratory was established in 2004 and has been headed by Prof. Jerzy Mozrzymas throughout its existence.

In earlier years of the former Division's operation, important research topics included issues related to theoretical biophysics, including the application of thermodynamics and statistical physics to describe transport across membranes and the regulation of cell homeostasis. Research on the interaction of proteins and low-molecular-weight substances with lipids was also one of the main scientific directions and made an important contribution to understanding the molecular basis of cell membrane function. In the 1970s and 1980s, spectroscopic and microcalorimetric studies of model membranes were developed, and the first electrophysiological test rig was established using the patch-clamp technique, which allows the recording of currents flowing through individual ion channels in membranes.
In the period from the 1980s to the mid-1990s, the individual's most significant scientific achievements can be considered to be: learning about some of the spectral properties, an important protein of the erythrocyte membrane skeleton, demonstrating the binding of glycolytic enzymes to lipid membranes, describing the modulation of acetylcholine receptors in states of physiology and pathology, characterising voltage-gated potassium channels in chondrocytes, as well as in T lymphocytes of patients suffering from chronic renal failure, elucidating the role of calcium ions in the accumulation of heavy metals by certain bacteria.

Under the editorship of Prof. S. Miękisz and A. Hendrich, PhD, a textbook titled "Selected Issues in Biophysics" was developed in 1998, the authors of which were awarded the Ministry of Health and Social Welfare's Team Award in 1999.

Starting from 1997, research topics in the Department of Biophysics have focused on issues related to drug resistance in cancer and fungal cells, synaptic transmission and its modulation in the central nervous system, and the function of potassium channels in T lymphocytes and their modulation by various factors.

The development of many of the Department's experimental methods and research facilities progressed systematically. New scientific laboratories were established: the Confocal Microscopy Laboratory, the Cell Culture Laboratory and the Molecular Biology Laboratory. The Fluorescence Spectroscopy and Microcalorimetry Laboratory was modernised. During this time, research on synaptic transmission in the central nervous system, led by Prof. Jerzy Mozrzymas, was developing very dynamically.

The Department of Biophysics has co-organised scientific congresses, both national and international.

Due to the rapid development of research into electrophysiological processes in the nervous system, a new research unit in the Department of Biophysics – the Independent Neuro System Biophysics Laboratory – was established. Its organiser was Prof. Jerzy Mozrzymas, who also became its head. The studio began its activities in its new premises at ul. Chałubińskiego 3a in 2004.

The Department of Biophysics has come a long way in the last half-century from a small Division without its own premises and scientific apparatus to a research and teaching facility, integrated into the world science network, conducting research in the molecular basis of medical science.
